Biological Activity
OSI-027 is an orally bioavailable mTOR (mammalian target of rapamycin) kinase inhibitor with anti-tumor activity. OSI-027 binds to and inhibits both the raptor-mTOR (TOR complex 1 or TORC1) and the rictor-mTOR (TOR complex 2 or TORC2) complexes of mTOR, which may result in tumor cell apoptosis and a decrease in tumor cell proliferation. OSI-027 is the second generation mTOR inhibitor. This dual TORC1 and TORC2 activity differentiates OSI-027 from rapamycin and the various related analogs, or rapalogs which molecules only inhibit TORC1 activity. This dual mTORC1/C2 inhibitor OSI-027 is currently in clinical trials.
